Vitamin K2-7 + Cholecalciferol + Combination

Tablet

This combination is useful in people having low blood calcium levels; who do not get enough calcium from their diets. Calcium is necessary for many normal functions of body, especially bone formation and maintenance. Low blood levels of calcium may cause bone disease. Thus this combination helps in maintance of calcium & healthy bones.
Calcium Citrate is calcium salt of citric acid. Calcium salt can be used in the prevention of calcium deficiency states. It is also used as an adjunct in the prevention of osteoporosis.
Methylcobalamin is the neurologically active form of the vitamin B12 and has strong effect on the brain and nervous system being also very important for cell growth and production. Methylcobalamin is used in variety of processes, including: production of red and white blood cell, the regeneration of RNA and DNA, synthesis of proteins. It is useful in vitamin B12 deficiency, psoriasis, pernicious anemia & atopic dermatitis.
Cholecalciferol is a fat soluble vitamin D3. Vitamin D is important for the absorption of calcium and phosphorus. Having the right amount of vitamin D, calcium, and phosphorus is important for building and keeping strong bones.
Vitamin K2-7 shown to have significant importance for optimal utilization of calcium in the body related to bone and cardiovascular health. Daily supplementation is expected to be beneficial for the bone building process as well as for cardiovascular health. Different forms of vitamin K2 are well documented and shown to be safe in numerous humans studies.

Mechanism

Calcitriol: The two known sites of action of Calcitriol are intestine and bone. Calcitriol may also act on the kidney and the parathyroid glands. Calcitriol is the most active known form of vitamin D3 in stimulating intestinal calcium transport. Calcitriol, in conjunction with parathyroid hormone, stimulates resorption of calcium in bones while in kidney; it increases the tubular reabsorption of calcium.
Methylcobalamin: as we age our body's ability to methylate declines, contributes to the aging process. Methylcobalamin functions as methyl donor. Folate is involved in neurotransmitter synthesis and critical enzymatic reactions throughout the body. By depleting excess homocystine, folate benefits cardiovascular health and nervous system function.
Cholecalciferol: It helps in absorption of calcium. The most important function of vitamin D3 is its effect on bone growth, density and structural integrity. This vitamin is needed for proper integration of calcium and phosphorous to make strong, healthy bones.
Vitamin K2-7: Vitamin K functions in the post-translational modification of a number of vitamin K-dependent proteins such as osteocalcin, a bone protein containing gamma- carboxyglutamic acid. This protein is involved in calcium uptake and bone mineralization.

Pharmacokinetics

  • Absorption: Vitamin K2-7 is absorbed in the small intestine with the help of dietary fats; Cholecalciferol (Vitamin D3) is absorbed efficiently in the intestine when taken with food.
  • Distribution: Vitamin K2-7 is transported via lipoproteins and accumulates in bone and vascular tissues; Vitamin D3 is widely distributed and stored in adipose tissue and muscle.
  • Metabolism: Vitamin K2-7 is metabolized in the liver to active menaquinone forms; Vitamin D3 is hydroxylated in the liver to calcidiol and further converted in the kidney to calcitriol (active form).
  • Elimination: Vitamin K2-7 is eliminated mainly via bile and feces; Vitamin D3 metabolites are excreted through bile and urine.
  • Half-life: Vitamin K2-7 has a relatively long half-life (~2–3 days); Vitamin D3 (calcidiol) has a half-life of ~2–3 weeks, allowing sustained activity.

Side Effects

  • Mild nausea, headache, or stomach upset may occur.
  • Use cautiously in patients with kidney or liver impairment.
  • Not recommended with anticoagulants (Vitamin K2 may interfere).

Contraindication

  • Hypersensitivity to any component of the formulation.
  • Patients with hypercalcemia or calcium metabolism disorders.
  • Severe renal impairment or nephrolithiasis (kidney stones).
  • Hemochromatosis or other iron overload conditions (due to folate/B12 interactions).

Interaction

  • Anticoagulants (Warfarin, Heparin): Vitamin K2 may reduce effectiveness.
  • Antacids & Proton Pump Inhibitors: May reduce calcium absorption.
  • Tetracycline & Quinolone Antibiotics: Calcium may interfere with absorption.
  • Phenytoin & Phenobarbital: May reduce Vitamin D3 effectiveness.
  • Alcohol (chronic use): May impair folate and B12 metabolism.

Warning and Precautions

  • Use under medical supervision, especially in patients with osteoporosis or cardiovascular disease.
  • Monitor serum calcium and Vitamin D levels during long-term therapy to avoid hypercalcemia.
  • Patients with renal impairment should use calcium supplements cautiously to prevent kidney stone formation.
  • Vitamin K2 may interfere with anticoagulant therapy; monitor INR levels closely if on warfarin or similar drugs.
  • Excessive Vitamin D3 intake may cause toxicity; avoid prolonged high-dose use without physician guidance.
  • Pregnant and lactating women should consult a healthcare provider before use.
  • Store in a cool, dry place, away from direct sunlight and moisture.
  • Keep out of reach of children.
